چکیده

We have raised an antiserum, designated aEnhb-1, to a portion of the mouse En-2 protein containing the homeodomain. The antiserum detects both the En-1 and En-2 proteins in mouse, chick and Xenopus embryos by Western blot analysis. Using whole-mount immunohistochemistry, combined in some cases with scanning electron microscopy, we have examined the distribution of the proteins in the early embryos of these species. The major features of expression were similar. The initial production of En protein occurred, just before or during the formation of the first somites, in a band of the anterior neural plate in the prospective mid/hindbrain region. Later in development En-1 protein accumulated in the ventral ectoderm of the developing mouse and chick limb buds, indicating that a dorsal-ventral polarity is present as soon as any limb bud swelling is apparent and that, at least in the mouse, this polarity is established independently of the apical ectodermal ridge. In all three species, aEnhb-1 bound to a subset of ventro-lateral differentiating neurons in the spinal cord and hindbrain and their pattern of birth in the mouse reflected the division of the hindbrain into rhombomeres. En-1 protein also accumulated in a lateral stripe of dermatome in the mouse and chick, indicating a dorsal-ventral subdivision of this tissue. The results show that En expression is a good marker for pattern formation in a variety of tissues and will be useful in experimental studies designed to characterize further these processes.

P-126: Stem Cell Factor Increases Blastocyst Formation in Mouse Two-Cell Embryo Culture

Background: It is demonstrated that c-Kit( receptor of stem cell factor) mRNA is expressed in late 2-cell stage to the expanded and hatched blastocyst and the stem cell factor (SCF) transcript is detected in the oviduct and uterus. The aim of this study was to investigate the effect of different doses of SCF on mouse 2-cell embryo development in vitro. Materials and Methods: 4-6 weeks old femal...

P-30: Developmental Capacity and Blastocyst Formation of Thawed Tetrahedral Versus Non-Tetrahedral 4-cell Stage Mouse Embryos After Vitrification

Background: It was reported in a literature that approximately one third of the 4-cell stage embryos did not exhibit a tetrahedral shape. Non-tetrahedral embryos showed a lower in vitro developmental potential than tetrahedral embryos. Recently vitrification technology has been widely employed for embryo cryopreservation. The objective of this study was to prove our hypothesis that vitrified - ...

P-128: The Effect of DNA Methyl Transferase1 Inhibitor (RG108) on DNA Methylation Pattern of Cloned Mouse Embryos

Background: In somatic cell nuclear transfer (SCNT) method of cloning, transferred nucleus should be dedifferentiated from differentiated state to embryonic state. Molecular analysis showed that the reprogramming in the transferred nucleus was incomplete and cloned embryos have epigenetic abnormalities such as high DNA methylations levels. Since methylation in pre-implantation embryos has a cri...

Effects of blastocyst artificial collapse prior to vitrification on hatching and ‎survival rates and the expression of klf4 gene in mouse embryos

Although the rate of blastocysts implantation of embryos is higher than previous stages but their survival rate is lower than them, which could be attributed to the completely filled blastocoel cavity with liquid and increased possibility of the formation of ice crystals. This liquid could prevent the penetration of cryoprotecting materials into the embryos. In this study, we reduced the volume...
